ABSTRACT

Objective:To investigate the clinical characteristics and pathogenic gene mutation of lateral meningocele syndrome(LMS).Methods:We retrospectively collected the clinical manifestations, laboratory examination, imaging examinations, and genetic analysis of a neonate with LMS which was diagnosed at the Department of Neonatology of the Second Affiliated Hospital of Wenzhou Medical University in May 2020. Relevant literature up to February 2021, retrieved from PubMed, OMIM, CNKI, Wanfang, and CQVIP database with the terms of "lateral meningocele syndrome", " NOTCH3", were reviewed to summarize the clinical characteristics, pathogenesis, and genetic etiology of this disease. Results:A full-term male newborn was admitted to our hospital due to feeding difficulty 7 d after birth. The clinical characteristics included hypotonia, dysphagia, hypertension, lateral spinal meningocele, craniofacial anomaly, and cryptorchidism. Abnormal spinal MRI and brainstem evoked potential were also observed. Whole exome sequencing revealed a heterozygous frameshift variation c.6667_6686del(p.Ala2223Profs*12) of NOTCH3 gene located in 19p13.12, which was not detected in the parents. Only 12 English literature were retrieved, with 17 patients from 15 pedigrees. Out of the 18 patients including the index case, 10 were genetically diagnosed as LMS. The age at diagnosis ranged from 15 d to 55 years. Regarding the clinical features, multiple lateral thoracolumbar spinal meningoceles (18/18) was the most common one, followed by retrognathia and low-set ears (16/18), eyelid ptosis and down slanting palpebral fissures (15/18), hypotonia (13/18), hypertension (11/18), developmental delay (9/18), mixed or conductive hearing loss (9/18), cardiovascular dysplasia (7/18), and cryptorchidism (7/10). A total of nine NOTCH3 gene variants were detected, all were heterozygous variants, including six frameshift and three nonsense variants. Conclusions:LMS is caused by NOTCH3 gene mutation with the clinical characteristics including multiple lateral thoracolumbar spinal meningoceles, craniofacial dysmorphisms, hypotonia, hypertension, developmental delay, difficulty in feeding, cryptorchidism, etc.

ABSTRACT

Objective@#To compare the effect of different orthokeratology lenses in controlling the progression of low myopia in children, and to provide a reference for exploring effective prevention measures for eyesight of children.@*Methods@#A total of 175 cases (350 eyes) aged 8-12 years old who were fitted with orthokeratology lenses were collected in this retrospective study. The differences in the changes of the axis length (AL) and the spherical equivalent refraction (SER) were analyzed after wearing different orthokeratology lenses for one year, and the relationship between the change of AL, SER and gender, age was also analyzed.@*Results@#In the Mouldway group, Alpha group, Lucid group and CRT group, the Median ( P 25 , P 75 ) of AL changes were 0.23 ( 0.12 , 0.41), 0.30 (0.17, 0.45), 0.35 (0.16, 0.41) and 0.33 (0.23, 0.41)mm, and there were no statistical significant difference between four groups ( Z =7.70, P >0.05); The Median ( P 25 , P 75 ) of SER changes were -0.31 (-1.00, 0.28), -0.38 ( -1.22 , 0.13), -0.25 (-0.84, 0.13) and -0.63 (-1.13, 0.25)D, and there were no statistical significant difference between four groups ( Z =2.15, P >0.05). The age had negative correlation with the change of AL ( r =-0.26, P <0.05), but has nothing to do with the change of SER ( r =0.10, P >0.05). There was no statistically significant difference in the change of AL ( Z =2.25, P > 0.05 ) and SER ( Z =-1.50, P >0.05) among children of different genders.@*Conclusion@#Different orthokeratology lenses have no differences in controlling the growth of the AL and changing the SER.

ABSTRACT

Objective:To screen anti-EGFR components from Aconitum soongaricum Stapf. and investigate their inhibitory effect on HEK293 cells and EGFR/HEK293 high expression cell lines. Methods:A two-dimensional online system (EGFR/CMC-HPLC-MS) was built to screen anti-EGFR active compounds from Aconitum soongaricum Stapf.;molecular docking assay was performed to determine the binding affinity of the components to EGFR;MTT was used to confirm the inhibition effect of the screened compounds on HEK293 cells and EGFR/HEK293 high expression cell lines.Results:The screening results showed that aconitine,12-epi-napellin and songorine were the active components acting on EGFR like gefitinib; molecular docking assay showed the targeting components acting on EGFR in the similar manner with gefitinib used as a positive control drug; MTT assay confirmed the screened components had inhibitory effects on HEK293 cells and EGFR/HEK293 high expression cell lines in a dose-dependent manner within the dose range of 0.4- 50.0 μmol·L-1. Conclusion:The screening results are apparently consistent with the pharmacological effect. The online EGFR/CMC-HPLC-MS method has a good application prospect in the rapid screening of the active compounds in the complex system of traditional Chinese medicines,and fully demonstrates such advantages of cell membrane chromatography as simple and rapid with high efficiency.

ABSTRACT

Objective To explore the prevalence of depressive disorder in the Mosuo ethnic minority in Ninglang district,Yunnan Province.Methods By stratified random sampling,1 121 Mosuo subjects aged 15 or above were selected and assessed by the MINI-international neuropsychiatric interview according to the Structured Clinical Interview for DSM-V-TR Axis I Disorders-Patient Edition for mental disorders.Results The standardized time-point prevalence of major depressive disorder in Mosuo nationality was 1.74%,and 1.69% (95 % CI =1.32%-2.15 %) in males and 1.77% (95 % CI =1.39%-2.15 %) in females.There was no statistically significant difference in the prevalence of major depressive disorder between males and females (x2 =0.051,P>0.05).The standardized time-point prevalence of dysthymic disorder in Mosuo nationality was 0.78%,and 0.66% (95% CI=0.54%-0.78%) in males and 0.88% (95% CI=0.74%-1.02%) in females (x2=1.232,P>0.05).Those aged 40-54 years old had the highest adjusted prevalence of depressive episodes(1.48% (95%CI=0.77%-2.18%)).Conclusion The prevalence of depressive disorder in Mosuo nationality is in a low level,and the middle-age Mosuo people has the highest time-point prevalence.

ABSTRACT

Objective To investigate the prevalence and related characteristics of alcohol dependence in the Pumi people of Ninglang area in Yunnan Province.Methods By stratified multistage cluster randomization,542 residents were interviewed by psychiatrists using the structural questionnaire MINI-International Neuropsychiatric Interview.Results The prevalence of drinking in the study were 37.3%,13.6% and 22.5% for the male,female and the total sample.There were significant differences of alcohol dependence between males and females (x2 =304.310,P<0.01) in which males were significantly higher than those in females.The current prevalence of alcohol dependence in Pumi people was 4.8%(95%CI=3.0%-6.6%),and standardized current prevalence was 3.3%.The current prevalence of alcohol dependence in males was 9.3%,which was significantly higher than that (2.1%) in females (x2 =14.613,P<0.01).The prevalence of alcohol dependence in the Pumi people was 6.1% in the 21-30 years old,and 8.6% in the 51-60 years old.There were one case of major depression,one case of panic disorder,and five cases of insomnia.Conclusion The prevalence of alcohol dependence in Pumi people of Ninglang areas is high.Alcohol dependence has become one of the most common mental disorders and the public health problem.It is necessary to carry out prevention research in the future.

ABSTRACT

Objective To investigate the expression of TRIM28 and p16 in esophageal squamous cell car-cinoma(ESCC)and explore the possible correlation with them and clinicopathological characteristics. Methods The expression level of TRIM28 and p16 were measured by immunohistochemistry S-P in 136 cases with ESCC and 37 cases with normal esophageal mucosa,selected from the First Affiliated Hospital of Hebei North University De-partment of Pathology.The relationship between them and the clinical-pathological features was also analyzed.The localization of TRIM28 and p16 protein in ESCC was detected by immunofluorescence. Results(1)The positive rates of TRIM28 and p16 in ESCC were 91.2%and 32.4%,respectively,whereas in normal esophageal mucosa the corresponding rates were 24% and 57%,respectively.(2)Immunofluorescence results showed that TRIM28 and p16 protein were all mainly distributed in the nucleus of ESCC.(3)The abnormal expression of TRIM28 and p16 protein were all related to the invasion depth,TNM staging and lymph node metastasis in ESCC(P < 0.05).(4) The expression of TRIM28 was negatively correlated to the expression of p16 in ESCC(r =-0.284,P = 0.001). Conclusions The abnormal expression of TRIM28 and p16 may have synergistic effect on the initiation and devel-opment of ESCC.Co-detection of the expression of them may be useful for diagnosis of ESCC and guiding the clini-cal therapy.

ABSTRACT

Objective To explore MSCT optimal examination time window for patients with occult rib fracture, and provide objective evidence for forensic injury. Methods Totally 105 cases with chest trauma were retrospectively analysed. They were examined in the first week after trauma and re-examined in different time windows by MSCT. The quantities of occult rib fractures in the first examination were compared to those in re-examinations. Results The quantities of occult rib fractures at different inspection time windows were mostly different. There was no statistically significant between the quantities of rib fractures in the first week and the second week, the first week and the sixth week later, the fourth week and the fifth to sixth weeks (P>0.05). There was statistically significant between the quantities of rib fractures in the first week and the thrid to sixth weeks, the second week and the third to sixth weeks, the third week and the fourth to sixth weeks (P<0.05). Conclusion The result of the study imply that the fourth to sixth week is the optimal time window of MSCT examination in the occult rib fracture.

ABSTRACT

Objective To study the expression and significance of tumor metastasis suppressor gene-1(TMSG1) in esophageal squamous cell carcinoma (ESCC) and EC109 cells.Methods Immunohistochemistry S-P method was used to examine the expression of TMSG-1 protein in 136 cases of ESCC and 37 cases of normal esophageal mucosa.We analyzed the relationship between TMSG-1 and clinicopathological data of ESCC patients.EC109 cells were treated with 3 μg/mL of cisplatin (CDDP) in vitro for 24 h (the intervention group) and the control group was set up at the same time.The proliferation-inhibitory capability was analyzed with MTT assay.RT-PCR was used to examine the expression of TMSG-1 in the intervention group and the control group.Results The positive rate of TMSG-1 in ESCC and normal esophageal mucosa was 52.2% (71/136) and 94.6% (35/37),respectively.The expression of TMSG-1 in ESCC was significantly lower than that in normal esophageal mucosa (P<0.05).The expression of TMSG-1 was related to TNM stage,differentiation degree and lymph node metastasis (P<0.05).After EC 109 cells were treated with CDDP for 24 h,the proliferation inhibition rate was increased significantly compared with the control group (P<0.01).RT-PCR results showed that the expression of TMSG-1 in the cells of the intervention group was significantly higher than that in the control group (P< 0.01).Conclusion The abnormal expression of TMSG-1 may play a role in the development and metastasis of ESCC.Examination of TMSG-1 may be useful for making diagnosis and guiding clinical therapy of ESCC.

ABSTRACT

Objective To discussion the clinical study of milrinone injectionon on acute heart failure with poor effect of digitalis and diuretics.Metheds 90 cases of acute heart failure with poor effect of digitalis and diuretics patients from September 2014 to May 2016 in our hospital were selected,according to the random number table method is divided into control group and study group,45 cases in each groups,the control group was treated with conventional heart failure intervention program,the study group given milrinone injectionon on the basis of control group,two groups were treated for seven days.Serum neurohormonal factors,inflammatory factors and blood pressure were measured before and after treatment,and the clinical efficacy and complications were compared between two groups.Results Compared with before treatment,serum NT-proBNP,NE and ET-1 levels were decreased in two groups,NO content increased,the levels of serum TNF-α,IL-6 and Hs-CRP were decreased,the levels of LVEF and CI were increased,the levels of LVEDD and CTR were decreased,the differences were statistically significant(P<0.05).Compared with the control group,the levels of serum NT-proBNP,NE and ET-1 in the study group were lower,NO were higher,the levels of TNF-α,IL-6 and Hs-CRP in serum were lower,LVEF and CI were higher,LVEDD and CTR were lower,the differences were statistically significant(P<0.05).The effective rate of the study group was 86.67%,which was significantly higher than that of the control group(66.67%,P<0.05).All the patients were followed up and no cases were reported.There was no significant difference in the incidence of adverse reactions between the two groups.Conclusion Milrinone injectionon has a significant effect on acute heart failure with poor effect of digitalis and diuretics,can significantly reduce the neurotrophic factor and inflammatory factors,improve heart function,promote prognosis.

ABSTRACT

Objective:To observe the relationship between carotid intima-media thickness (CIMT) and hemoglobin A1c (HbA1c) control in type 2 diabetes mellitus (T2DM) patients.Methods:328 cases with T2DM were selected from the endocrinology department of our hospital.All patients underwent assessment,biochemical test and CIMT measurement.According to the value of CIMT,patients were divided into normal CIMT group (<0.9 mm) and abnormal CIMT group (>0.9 mm).A multi-factor Logistic regression was performed to analyze the risk factors associated with the abnormal CIMT.Results:(1) Among the 328 T2DM patients,154 cases had normal CIMT and 174 cases had abnormal CIMT.(2) Pearson correlation analysis indicated that the cholesterol and HbA1c levels were positively correlated with CIMT (P< 0.05).(3) Univariate analysis showed that the incidence of CIMT was associated with age (t=4.132,P=0.041),systolic blood pressure (t=8.456,P<0.01),HbA1c≥ 9.0% (x2=9.912,P<0.01),cholesterol 0=5.549,P=0.018),triglyceride (t=6.592,P=0.008),UA(t=9.618,P<0.01),fasting plasma glucose (t=4.592,P=0.037).(4) Multi-factor Logistic regression indicated that age,systolic blood pressure,cholesterol and HbA1c≥ 9.0% were the independent risk factors for the onset of CIMT (P <0.05).Conclusions:The level of HbA1c in T2DM patients was associated with CIMT;And HbA1c≥ 9% was the independent risk factors for the increase of CIMT.

ABSTRACT

Objective To explore the influence of carvedilol combined with diuretics on BNP and cardiac function of patients with mild to moderate cardiac insufficiency.Methods87 cases of patients with mild to moderate cardiac insufficiency treated in Zhejiang Greentown Cardiovascular Hospital from June 2014 to June 2016 were divided into the groups randomly.The control group of 44 cases were given carvedilol treatment, and the study group were combined with spironolactone treatment on the basis of the control group, with the total course for a month.Clinical effects were contrasted after a month of treatment in the two groups, in order to explore the influence of carvedilol combined with diuretics on BNP and cardiac function of patients with mild to moderate cardiac insufficiency.ResultsBefore treatment, there was no significant difference in serum BNP level between the control group and the study group.After treatment, serum BNP level significantly decreased when compared with those before treatment in the two groups (P< 0.05), which of the study group was significantly lower than that in the control group(P< 0.05).Before treatment, there were no significant differences in the levels of cardiac function indexes, and after treatment, heart rate, EF (%), FS (%), LVEDD (mm) the four indicators significantly improved when compared with those before treatment, and heart rate, LVEDD (mm) levels were significantly lower than those in the control group(P<0.05), and EF (%), FS (%) levels were significantly higher thatn those in the control group(P<0.05).After treatment, the significant effective rate (73.91%) and total effective rate (95.35%) in the study group were significantly higher than that of (56.82%) and(86.36%) in the control group (P<0.05).After treatment, body function, life satisfaction, mental health, social adjustment the four parts of quality of life index scores were significantly higher than those in the control group(P<0.05).ConclusionIt has obvious improvement effect of carvedilol combined with diuretics on BNP and cardiac function of patients with mild to moderate cardiac insufficiency.It is worthy of clinical application.

Objective To survey the incidence of adverse reaction induced by traditional Chinese medicine injection and provide valuable suggestion for using traditional Chinese medicine injection.Methods Clinical data of 1860 patients who were under treatment of traditional Chinese medicine injection during January 2010 to December 2014 were studied to survey adverse reaction incidence, effected system-organs and risk factors of adverse reaction.Results Comparison of incidence adverse reaction of traditional Chinese medicine injection, Qingkailing injection adverse occurrence rate reached the highest 11.76% for all types, followed by Shenmai injection and Shuanghuanglian injection.Adverse reactions involving the organ and the performance was compared, adverse reactions mainly involved the organs for the skin and accessories, circulatory system, accounting for 56.15%, the incidence of adverse reactions of the rest of the organ system was less.Not determine the treatment based on differentiation of symptoms and signs, combined with injection, ultra treatment application and distribution of liquid static for a long time all differences were statistically significant ( P <0.05), leading to the main risk factors of the occurrence of adverse reactions.Conclusion Traditional Chinese medicine injections occurred and the incidence of adverse reactions was higher, wider range, organ involvement mainly skin and its appendages and circulatory system, not dialectical treatment and rational drug use is lead to adverse risk factors, actively preventment and timely treatment to reduce the incidence of adverse reactions.

ABSTRACT

Objective To explore the effect of telephone intervention on discharged patients after total hip replacement. Methods 214 participants were assigned to the intervention group including 104cases receiving 6 months of telephone intervention and the control group including 110 cases receiving routine nursing by random number table method. The intervention were taken at 1,2 week and 1,2,3,6 month after patients discharged. The outcome measurements were the self-designed questionnaire of functional exercise compliance and Harris scale at 1, 3, 6 month patients discharged. Results The scores of self-designed questionnaire of functional exercise compliance were (42.24±4.58), (38.55±5.84), (36.98±5.41) at 1, 3, 6 month discharge from hospital in the intervention group, while in the control group was (38.79 ± 5.98), (35.45 ± 6.44), (32.45 ± 6.04) respectively. There were significant between two groups, t value was 3.290, 2.567 and 4.024 respectiveliy, P<0.05. There was no significant of Harris scale′s scores between two groups when they discharged. The scores of Harris scale were (66.51±6.64), (75.30± 6.65), (89.44±4.98) at 1, 3, 6 month discharge from hospital in the intervention group, while in the control group was (61.31 ± 6.68), (70.56 ± 8.01), (81.18 ± 6.13) respectively, and it has significant between two groups, t value was 4.232, 3.509 and 8.076, P<0.05. Besides, the scores of self-designed questionnaire of functional exercise compliance were decline and the scores of Harris scale were up along the time patients discharged in two groups(P < 0.05). Conclusions Telephone as a simple, convenient, efficient tool could enhance patients functional exercise compliance and hip function after total hip replacement.

Objective To investigate the relationship of RhoA and Snail expressions, and the invasion and metastasis in salivary adenoid cystic carcinoma (SACC). Methods The expressions of RhoA protein and Snail protein in 55 samples of SACC (SACC group ) and 20 samples of para-carcinoma normal tissues(control group) were detected using immunohisto?chemical method. The relationship between RhoA protein and Snail protein expressions and clinical and pathological charac?teristics were analyzed. Results The positive expressions of RhoA protein (69.1% vs 5.0%) and Snail protein (72.7% vs 10.0%) were significantly higher in SACC group than those in control group (P < 0.05). The positive expression rates of RhoA protein and Snail protein were significantly higher in patients with lymph node metastasis than those in patients with?out lymph node metastasis. The positive expression rates of RhoA protein and Snail protein were significantly higher in pa?tients atⅢ+Ⅳstage than those in patients atⅠ+Ⅱstage. The positive expression rates of RhoA protein and Snail protein were significantly higher in substantive carcinal tissues than those in screen roller type and tubular carcinal tissues. The posi?tive expression of Snail protein was significantly higher in substantive and tubular carcinal tissues than that in screen roller type carcinal tissues (P<0.05). There were no significant differences in positive expression rates of RhoA and Snail between different gender, age and different carcinal tissues. There was a positive correlation beween expression rates of RhoA and Snail protein in SACC (r=0.414, P<0.001). Conclusion RhoA and Snail may both facilitate the infiltration and metastasis of SACC through RhoA/ROCK/PKD1/NF-kappa B/Snail signaling pathways.

ABSTRACT

90 extracted single rooted human teeth were inoculated with E.faecalis.The cannals of the teeth were irrigated with MTAD, 5.25% NaClO,compound chlorhexidine gargle,tinidazole mouthwash,distilled H2 O(infection control)and autoclave followed by distilled H2 O(autoclaving control),respectively(n =1 5).Then E.faecalis infection of the samples were examined.The results showed that MTAD had significantly greater antibacterial effects than 5.25% NaClO,tinidazole mouthwashes,compound chlorhexidine gargle and tinidazole mouthwash(P <0.05).There was no significant difference between each 2 of the other 3 irrigants.

OBJECTIVE@#To investigate the expression and clinical significance of OPN, CD44v6 and MMP-9 in laryngeal squamous cell carcinomas (LSCC).@*METHOD@#The expression of OPN, CD44v6 and MMP-9 in the 47 cases of LSCC and 10 cases normal mucosa were detected by immunohistochemical method. The correlation between OPN, CD44v6 and MMP-9 proteins expressions and the correlation between them and clinicopathological feature were also studied.@*RESULT@#(1) The positive expression rates of OPN, CD44v6 and MMP-9 were 63.8%, 76.6% and 68.1% in LSCC of 47 cases and, and 10%, 30% and 0 in the tumor adjacent issue. (2) The positive rate of OPN and CD44v6 was significantly correlated with the clinical staging, the pathological grading and the metastasis of cervical lymph node (P 0.05). (3) The expression of OPN was well correlated with the positive rate of CD44v6 and MMP-9 in LSCC (r = 0.421, P < 0.01, r = 0.340, P < 0.05). The expression of positive rate of CD44v6 and MMP-9 was well related in LSCC as well (r = 0.376, P < 0.01).@*CONCLUSION@#There is a high level of expression of OPN, CD44v6 and MMP-9 in LSCC. OPN, CD44v6 and MMP-9 are positively related and involved in the invasion and metastasis of LSCC.

Objective To investigate the possible mechanisms of ox-LDL on vascular smooth muscle cells (VSMC) with and without exogenous zinc finger protein A20 gene. Methods VSMC were trans-fected with plamid containing A20 gene and incubated with ox-LDL. TLR4 and LOX-1 mRNA expression were measured by RT-PCR. Results After VSMC incubated with ox-LDL, the expression of LOX-1 and TLR-4 mRNA reached a higher level. However, when VSMC were transfected with plasmid containing A20 gene, the NF-kB nuclear translocation and the ability of migration reduced to normal level. Conclusions ox-LDL can significantly activate NF-kB signaling system and enhance migration in VSMC. The activation may induce the inflammatory response in arterial wall, the migration of VSMC to the intima, and triggering the process of atherosclerosis. Transfecting plasmid containing A20 gene significantly inhibited the activation of NF-kB and the ability of migration of VSMC. A20 effects might be through inhibiting NF-kB signaling. Meanwhile, A20 blocked the development of atherosclerosis.

ABSTRACT

Objective To explore the influence of studying in the elderly university on the cognitive function for elderly. Methods Elderly university group (EUG) was consised of 105 subjects aged ≥60 who studied in the elderly university for one year. Controlled group(CG) was consisted of 105 elderly over 60 years of age who were siblings for subjects and would not studied in elderly university. Meanwhile, it was excluded if they were working. Two groups were excluded depression patients before study. The assessment tools were composed of Center for epidemiological survey of depression scale (CES-D), activity of daily living scale(ADL), mini-mental state examination (MMSE)and Wechsler's adult intelligence scale(WAIS). There were no significant statistics difference in scales of CES-D, ADL, MMSE and WAIS between two groups. Results There were no significant statistics difference of CES-D, ADL and MMSE between two groups after a year, however, the scales of ADL for controlled group after one year was higher than that before one year( t = 2. 191, P = 0.031 ). The MMSE mark shows one(0.95% )sustain cognitive impairment in EUG after one year; meanwhile there were six elders of (5.77%)come up to critical value of cognitive impairment in CG, the difference was almost statistical significance (P=0. 058 ). The WAIS mark demonstrated the total scale, VQ scale and PQ scale was declining. The score of total scale and perform scale of WAIS in CG were (110.3 ±8.2 )and (110.9 ±7.7) respectively. The score of total scale and perform scale of WAIS in CG were ( 107.8 ± 9.6 ) and ( 107.9 ± 13.4 ) respectively. There were significant difference between two groups(P=0. 044, P=0. 049). Furthermore, after one year the score of total scale of WAIS in CG( 110. 6 ± 7.4 ) outstanding be lower than that one year ago(P = 0. 022 ). There was no significant means for the total scale , verbal scale and perform scale of WAIS in CEG before and after year.Conclusion The program of continue education may play a positive role to stay or to improve the descending of the cognitive function in the elderly.

ABSTRACT

Aim To investigate the effects of 5-HT_4 receptor agonist and 5-HT_3 receptor antagonist 2-[1-(4-piperonyl)piperazinyl]benzothiazole on rat heart rhythm and the involved ionic mechanisms.Methods Langendorff-perfused rat hearts were subjected to 0.1~10 μmol·L~(-1) 2-[1-(4-piperonyl)-piperazinyl]benzothiazole for 15 minutes with simultaneous ECGs recording.The whole-cell patch-clamp electrophysiology was used to record effects of 2-[1-(4-piperonyl)piperazinyl]benzothiazole on inward rectifier K~+ current(I_(K1)),transient outward K~+ current(I_(to)),resting membrane potential(RMP)and action potential(AP)in enzymatic dissociated rat ventricular myocytes.Results In ex vivo Langendorff-perfused hearts,0.1~10 μmol·L~(-1) 2-[1-(4-piperonyl)piperazinyl]benzothiazole elicited singnificant rhythm disturbances.In the presence of 10 μmol·L~(-1) agent,the total of PVB were 236±37,87.5%(7/8)hearts exhibited VT,and 62.5%(5/8)hearts exhibited VF(P<0.01).At the concentration of 0.1~10 μmol·L~(-1),2-[1-(4-piperonyl)piperazinyl]benzothiazole could inhibit I_(K1)(EC50=0.74 μmol·L~(-1))and I_(to)(EC50=2.16 μmol·L~(-1)),decrease RMP and prolong action potential duration(APD)in concentration-dependent manners(n=6,P<0.01).Conclusion Inhibition of IK1,Ito and resultant prolongation of APD,depolarization of RMP might be the critical causes for induction of arrhythmias by 2-[1-(4-piperonyl)piperazinyl]benzothiazole in rat.

ABSTRACT

Objective To observe the morphologic changes of pancreatic islets in diabetic rats and the protective effects of rosiglitazone natrium (RSG) on the remnant islet.Methods 8 rats were randomly selected from 68 male Sprague-Dawley rats for normal control group (group A),the rest were administered with STZ,and those with FBG≥16.7mmol/L were served as diabetic models,then the rats were randomly assigned as RSG-treated group (group B,n=24).RSG were applied via intragastric administration at a dose of 5mg/(kg·d) in group B.72 hours after the administration of STZ and 1-,2-,4 and 7 week after the administration of RSG,4 rats were randomly selected respectively,pancreas specimens were obtained and preserved.The morphologie changes were observed under light microscopes and transmission electron microscope.Results The diabetic symptoms of the rats in groupe B lessened significantly,and the FBG levels were decreased gradually,the morphologic injuries of remnant islet were recovering. The amount of secretory granule in beta cells increased gradually.Conclusion Rosiglitazone natrium call attenuate morphologic injuries of the islets and protect the remnant islet in type 1 diabetes mellilus rats.
